2.2 抗肿瘤活性采用SRB法并结合显微镜检测细胞形态学变化,测定了化合物1~8对人慢性髓原白血病细胞K562肿瘤细胞的抑制活性,结果表明环二肽类化合物4、5和7在10μmol/L时即能观查到细胞坏死活性,抑制率分别为15.4%、19.3%和18.6%,其他化合物在100μmol/L高浓度时也未检测到相关活性。
3 讨论
本文采用活性追踪的方法从一株海洋放线菌S1001的活性部位分离鉴定了1个异黄酮类化合物,1个苯甲酸类衍生物,以及6个环二肽类化合物,并经活性测试阐明了环二肽类化合物为该菌株的主要活性相关成分。文献报道也显示环二肽类化合物可作为免疫功能调节剂和抗肿瘤制剂,并且体内体外试验证明均有效[9,10],值得进一步研究。菌株S1001的次级代谢产物比较单一,主要为环二肽类化合物且产量较大,因此可以考虑作为环二肽的生产菌株加以利用。
