Abstract
Capillary electrophoresis with Ru(bpy)3
2+ electrochemiluminescence detection was established to the
determination of the alkaloids in henbane. Parameters related to the injection,separation and detection were
optimized. Under the optimized conditions, ECL detection at 1.2 V, 50 mmol/L phosphate buffer at pH 7.48 in the
detection cell, phosphate buffer at pH 8.48 as running buffer, injection voltage 12 kV, injection time 9 s, the
standard curves were linear between 0.29 and 5.8 mg/L for atropine and between 3.0 and 303 mg/L for
scopolamine, respectively. Detection limits of 0.014 mg/L for atropine and 0.03 mg/L for scopolamine were
obtained. Compared with traditional analytical method, the present method displayed advantages of sensitivity,
selectivity, linearity and small sample consumption. Due to an advanced anti-jamming capability of ECL detection,
a satisfactory result was achieved for the detection of drugs in herbs containing a large amount of coexistent
substances. Developed method was successfully applied to determine the alkaloids in henbane.
