【摘要】 目的分离和鉴定白附子炮制后新增化学成分。方法采用高效液相色谱(HPLC)法确定提取溶剂,用硅胶柱和制备高效液相分离纯化,硅胶G薄层板和HPLC检识,通过理化性质和光谱数据确定化合物结构。结果新产生的化学成分为5-羟甲基糠醛(Ⅰ),还分离到了其结构类似物质双[5-甲酰基-糠基]-醚(Ⅱ)。结论首次从制白附子中分离得到5-羟甲基糠醛和双[5-甲酰基-糠基]-醚,为中医临床炮制用药提供依据。

Isolation and Identification of the Newly Added Chemical Constituent from Processed Rhizoma Typhonii

Abstract:ObjectiveTo study the newly added chemical constituent from processed Rhizoma Typhonii.MethodsThe extraction solvent was determined by HPLC. The constituents were isolated and purified by column chromatography and preparative HPLC method and determined by TLC and HPLC. The structures were elucidated based on chemical and spectral analysis. ResultsTwo compounds were isolated from the chloroform extracts. Their structures were determined as 5-hydroxymethyl-2-furaldehyde (Ⅰ) and bis(5-formylfurfuryl)ether(Ⅱ).Conclusion5-hydroxymethyl-2-furaldehyde and bis(5-formylfurfuryl)ether were isolated from processed Rhizoma Typhonii for the first time.

白附子(习称禹白附)为天南星科犁头尖属植物独角莲Typhonium giganteum Engl.的干燥块茎,辛,温;有毒。归胃、肝经,具有祛风痰、定惊搐、解毒散结止痛之功效[1]。白附子有毒,多用其炮制品。《中国药典》(2005年版)载制白附子采用姜矾制。从白附子炮制前后的高效液相指纹图谱对比中,发现制白附子指纹图谱中有新的色谱峰产生,说明白附子炮制后产生了新的化学成分。本研究从制白附子中提取、分离炮制后新产生的成分,并进行结构鉴定。
